چکیده

De Meio and Lewycka (1) have obtained indirect evidence that conjugation of dehydroisoandrosterone with sulfuric acid occurs in a system consisting of microsome-free supernatant fluid of rat liver, adenosine triphosphate (ATP), and magnesium and sulfate ions. They showed that, following incubation, some of the Zimmermann-positive material remains in the aqueous phase after ether extraction at pH 7, but is recovered by subsequent extraction with n-butanol. We wish to present more direct evidence that a number of steroids are conjugated with sulfuric acid in such a system while others are not.

Steroid sulfatase and estrogen sulfotransferase in human endometrial carcinoma.

PURPOSE Intratumoral metabolism and synthesis of estrogens are considered to play important roles in the pathogenesis and/or development of human endometrial carcinoma. Steroid sulfatase hydrolyzes biologically inactive estrogen sulfates to active estrogens, whereas estrogen sulfotransferase sulfonates estrogens to estrogen sulfates. However, the status of steroid sulfatase and/or estrogen sulf...

The conjugated steroids. IV. The hydrolysis of ketosteroid sulfates.

Specific hydrolysis of the separate groups of steroid conjugates appeared to offer the simplest technique for achieving the hydrolysis without chemical modification of the steroids (3-5). A partial solution to this problem was offered when it was found that steroid glucuronides could be hydrolyzed specifically by the use of /3-glucuronidase concentrates of either animal (Cohen (3)) or bacterial...

In Vivo Regulation of Steroid Hormones by the Chst10 Sulfotransferase in Mouse*

Chst10 adds sulfate to glucuronic acid to form a carbohydrate antigen, HNK-1, in glycoproteins and glycolipids. To determine the role of Chst10 in vivo, we generated systemic Chst10-deficient mutant mice. Although Chst10(-/-) mice were born and grew to adulthood with no gross defects, they were subfertile. Uteri from Chst10(-/-) females at the pro-estrus stage were larger than those from wild-t...

Biosynthesis of Steroid Sulfates by Human Ovarian Tissue.

Sulfate esters of steroid hormones had been considered to represent end products of steroid metabolism designed for excretion. Sulfation of steroids in vitro had been demonstrated with mammalian liver preparations, and it had been generally assumed that the liver was the major site of production of these compounds in man. Revision of this view became necessary with the demonstration that the st...

Steroid sulfatase: a pivotal player in estrogen synthesis and metabolism.

Steroid sulfatase plays a pivotal role in regulating the formation of biologically active steroids from inactive steroid sulfates. It is responsible for the hydrolysis of estrone sulfate and dehydroepiandrosterone sulfate to estrone and dehydroepiandrosterone, respectively, both of which can be subsequently reduced to steroids with estrogenic properties (i.e. estradiol and androstenediol) that ...
